These products, also referred to as skin-whiteners, brighteners, or bleach creams, try to even and lessen skin coloration due to the formation of excessive melanin.<\/p>\n<h4><strong>The Biology of Skin Pigmentation<\/strong><\/h4>\n<p>The skin pigmentation process is complex. It\u2019s believed there are at least 14 primary mechanisms involved in the formation of skin color, as well as 3 secondary ones. Skin color originates in the epidermis, where the pigment-producing cells, melanocytes, are located.<\/p>\n<p>Upon UV exposure, these cells produce pigment, called melanin. This process is referred to as melanogenesis (see figure below-right) and is controlled by the copper-containing enzyme tyrosinase. Tyrosinase is located in vesicles called melanosomes which are inside melanocytes.<\/p>\n<p>Two types of pigments are synthesized in melanosomes: eumelanin and pheomelanin. New actives have been developed that claim to interfere with the transfer of melanosomes to keratinocytes. Another efficient mechanism would be to block the up-regulation of tyrosinase and block the stimulation of melanocyte dendricity. This could be achieved by interfering with the binding of keratinocyte mediators to their melanocyte receptors or by inhibiting the production of these mediators.<\/p>\n<h4><strong>Skin-Lightening Strategies<\/strong><\/h4>\n<p>Skin-lightening strategies include utilizing combinations of materials that work by different mechanisms. All skin-lightening formulations should contain a <a href=\"http:\/\/www.ulprospector.com\/en\/na\/PersonalCare\/search?k=chelating+agent&amp;st=31\" target=\"_blank\" rel=\"noopener\">chelating agent <\/a>(<a href=\"http:\/\/www.ulprospector.com\/en\/eu\/PersonalCare\/search?k=Chelating+Agent&amp;st=31\" target=\"_blank\" rel=\"noopener\">EU<\/a>), an <a href=\"http:\/\/www.ulprospector.com\/en\/na\/PersonalCare\/Product\/search?k=Antioxidants&amp;sug=1&amp;st=31\" target=\"_blank\" rel=\"noopener\">antioxidant <\/a>(<a href=\"http:\/\/www.ulprospector.com\/en\/eu\/PersonalCare\/Product\/search?k=Antioxidants&amp;sug=1&amp;st=31\" target=\"_blank\" rel=\"noopener\">EU<\/a>), and a <a href=\"http:\/\/www.ulprospector.com\/en\/na\/PersonalCare\/search?k=skin+soothing+agent&amp;st=31\" target=\"_blank\" rel=\"noopener\">skin-soothing agent <\/a>(<a href=\"http:\/\/www.ulprospector.com\/en\/eu\/PersonalCare\/search?k=Skin-Soothing+Agent&amp;st=31\" target=\"_blank\" rel=\"noopener\">EU<\/a>).<\/p>\n<h4><strong>Enzyme Inhibition<\/strong><\/h4>\n<ul>\n<li>Tyrosinase synthesis and activity.<\/li>\n<li>Phenylalanine hydroxylase (PAH).<\/li>\n<li>Tyrosinase hydroxylase isoenzyme 1 (TH-I).<\/li>\n<li>Dopa oxidase<\/li>\n<li>Tyrosinase-related proteins (TRP-1,2) similar to tyrosinase.<\/li>\n<li>Antioxidant enzymes such as catalase, glutathione peroxidase, glutathione reductase and thioredoxin reductase.<\/li>\n<li>Protease-activated receptor 2 (PAR-2) inhibition may have an impact on melanosome transfer.<\/li>\n<li>Binding to the peroxisome proliferator activated receptor-gamma (PPAR-gamma) and thereby inhibiting the mRNA and protein levels.<\/li>\n<\/ul>\n<h4><strong>Other Mechanisms<\/strong><\/h4>\n<ul>\n<li>Inhibit melanocyte dendricity.<\/li>\n<li>Inhibit melanin stimulating hormone (MSH).<\/li>\n<li>Melanosome transfer inhibitors.<\/li>\n<li>Reduce skin irritation, which is known to stimulate hyperpigmentation.<\/li>\n<li>Inhibit microphthalmia-associated transcription factor gene (MITF) expression. MITF plays a central role in melanin synthesis, as well as melanosome biogenesis and transport.<\/li>\n<li>Increase cell turnover and desquamation.<\/li>\n<li>Chelating agents &#8211; <a href=\"http:\/\/www.ulprospector.com\/en\/na\/PersonalCare\/search?k=copper&amp;st=31\" target=\"_blank\" rel=\"noopener\">copper<\/a> (<a href=\"http:\/\/www.ulprospector.com\/en\/eu\/PersonalCare\/search?k=copper&amp;st=31\" target=\"_blank\" rel=\"noopener\">EU<\/a>) is essential for tyrosinase activity. Utilize antioxidants.
